Simple answer: use them as you need to!

That message means that VPX has found 41 plug-ins that it can use. If you go to Effects>Extra Effects>Plug ins and, if you have a video object highlighted on the timeline, a screen like this will open;

Simply click on any of those available and choose to Insert Plug in; it will now be available to use with that video object.
