How can I synthesise my dog's bark?

OK -I am new to all this. I can record audio and midi sources and mix them. Is it possible to record an audio sample and then use it as a "sound" such that it can be played at any pitch (like the Fairlight CMI all those years ago)?

What you're asking about is called a "sampler".

 

I haven't tried it, but you can probably do what you want by installing one of these software samplers in Samplitude as a VST instrument.
